Nonspecific background staining can be attributed to endogenous avidin-biotin, to nonspecific antibody binding and to the presence of endogenous enzymes. To enhance staining, reduce nonspecific staining and avoid false-positive staining results, blocking methods are required in all IHC staining techniques. Blocking methods include those that utilize unlabeled avidin-biotin, serum and enzyme-quenching chemicals.
